ICAT and ITRAQ Labeling with Mass Spectrometry
- ICAT and ITRAQ labeling with mass spectrometry are approaches to compare proteins from 2-4 different states or conditions. Proteins are determined by ms/ms analysis and ICAT (cysteine label) or ITRAQ (amine label) give results to determine relative quantities. Numbers of proteins identified depends on the complexity of the sample and up front separation prior to mass spectrometry.
- Our facility is equipped to apply these approaches to purified or unprocessed samples including intact tissue, cell culture, dilute and concentrated solution mixtures. The only requirement is sufficient starting material to achieve 100 mg/sample.
ICAT - Method to label cysteines for protein relative abundance quantitation
ITRAQ - Method to label primary amines for protein relative abundance quantitation
ITRAQ Application Note - example of multiplex protein quantitation using iTRAQ TM Reagents
