Glucokinase Regulatory ProteinGlucokinase regulatory protein (GKRP) has the molecular weight about 70 kDa and recently it was shown to inhibit the GK enzyme activity by acting at a specific allosteric binding site in hepatic GK. GKRP and GK are localized in the nucleus of hepatocytes but only GK is translocated to the cytoplasm in response to high glucose concentrations. This correlation between glucose metabolism and translocation of GK from nucleus to cytosol is consistent with the known kinetic properties of GK in association with GKRP. It has been shown that GKRP transduces the metabolic effects of fructose-6-phosphate and fructose-1-phosphate by modulating the interaction of GK and GKRP. |
