Growth Curves

Start a 5ml culture in selective media and grow to saturation (overnight)
Subculture to 20ml selective media in a Klett flask
- 200µ l saturated cells
- 20ml media
Incubate at desired temp with shaking
Take Klett readings at appropriate time points (every hour during log phase)
- Klett meter instructions
- Turn power and light switches on
- Zeroing:
  - Put in flask with just the media you’re using
  - Turn top knob until the line matches the arrowhead
Reading:
- Start with zero reading
- Put in flask with culture
- Turn big knob until arrowhead lines up with the black line
- Read Klett Units
