The Ohlendorf lab is interested in how protein structure leads to function. To this end, x-ray crystallography is used to determine the positions of atoms in space. The structure is then analyzed in light of the biochemical properties of the protein, suggesting mechanistic roles for key residues. These hypotheses are tested by producing mutant proteins and measuring changes in their function and structure.
