Extradiol Dioxygenases

Homoprotocatechuate 2,3-dioxygenase (HPCD) catalyzes the cleavage of homoprotocatechuate (3,4-dihydroxylphenyl acetate). In Brevibacterium fuscum HPCD uses Fe(II) as a cofactor; in Arthrobacter globiformis CM-2 HPCD uses Mn(II) as a cofactor. The structures of HPCD from both organisms has recently been determined by the Ohlendorf lab. The amino acid sequences of these HPCD's are 81% identical

Work on HPCD is in collaboration with Dr. John Lipscomb and Dr. Larry Wackett of the Dept. of Biochemistry.
